**NBIP-1968** is an investigational long-acting triple receptor agonist that entered first-in-human testing in August 2026. Neurocrine Biosciences designed the molecule to engage the GLP-1, GIP and glucagon receptors from a single peptide backbone, and it is one of the few metabolic candidates disclosed this year with no human data of any kind behind it yet.

Development stage

NBIP-1968

- Preclinical
- Phase 1
- Phase 2
- Phase 3

- Approved

What has been shown in humans?

Nothing yet. The Phase 1 study opened in August 2026 and is the first time the compound has been administered to people. No results have been reported, and the preclinical package behind it has not appeared in a peer-reviewed journal. Findings in animals do not establish effects in humans.

Discovered internally by Neurocrine Biosciences. The amino acid sequence has not been disclosed and no CAS number is in circulation. The sponsor has described it as intended for eventual fixed-combination use with NBIP-2118, a separate corticotropin-releasing factor type 2 receptor agonist also in Phase 1.

Portal to Wellness does not sell this compound.

## What was actually announced

Neurocrine Biosciences [announced in August 2026](https://neurocrine.gcs-web.com/news-releases/news-release-details/neurocrine-biosciences-announces-initiation-phase-1-clinical-6) that it had begun a Phase 1 clinical study of NBIP-1968 in adults across a range of body mass index categories. The stated objective of the first stage is safety, tolerability and pharmacokinetics in ascending-exposure cohorts — the standard opening question of any first-in-human programme, and the only question a Phase 1 of this design is built to answer.

The announcement was also covered by [Clinical Trials Arena](https://www.clinicaltrialsarena.com/news/neurocrine-begins-nbip-1968-phase-i-trial/) and [BioPharm International](https://www.biopharminternational.com/view/neurocrine-biosciences-begins-phase-1-study-of-glp-1-gip-glucagon-triple-agonist-nbip--1968-for-obesity) . What none of the coverage contains is a sequence, a structure, or a number from a human being. That absence is the single most useful fact about this compound right now.

## The glucagon arm is the part worth watching

Triple agonism at GLP-1, GIP and glucagon receptors is not new as a concept — retatrutide has been the reference molecule for that receptor combination for several years. What Neurocrine has emphasised in its own description is that the glucagon receptor component was deliberately balanced rather than maximised, on the reasoning that glucagon receptor engagement is where tolerability problems have historically appeared in this class.

That is a design assertion, not a finding. Whether the balance chosen actually behaves differently in people is precisely what has not been measured. Researchers following this class should treat the “balanced glucagon activity” framing as a hypothesis the Phase 1 exists to test, and not as an established property of the molecule.

## Where NBIP-1968 sits against compounds that do have human data

| Compound | Receptor targets | Stage | Human data published? |
| --- | --- | --- | --- |
| **NBIP-1968** | GLP-1 / GIP / glucagon | Phase 1, opened Aug 2026 | None |
| Retatrutide | GLP-1 / GIP / glucagon | Phase 3 | Yes, multiple readouts |
| [Tirzepatide](https://portaltowellness.net/shop/tirzepatide-15mg/) | GLP-1 / GIP | Approved (as a prescription medicine) | Yes, extensive |
| MBX 4291 | GLP-1 / GIP co-agonist prodrug | Phase 1 | Blinded interim only |
| HM17321 | CRFR2 (urocortin-2 analog) | Phase 1 | None; animal work only |
| NBIP-2118 | CRF2 receptor agonist | Phase 1 | None |

The table makes a point worth sitting with. Two of the six rows are compounds whose receptor target — CRFR2 — was, until recently, represented in the obesity pipeline by a single programme. Neurocrine disclosing NBIP-2118 alongside NBIP-1968 means the corticotropin-releasing factor route now has at least two sponsors behind it, which is usually the point at which a mechanism stops being one company’s idiosyncratic bet. We covered the other one in [our write-up of HM17321](https://portaltowellness.net/research/hm17321-muscle-preservation-obesity-research/) .

## What has not been disclosed

Four things, all of them material:

- **The sequence.** No primary structure has been published. Without it there is no route to independent characterisation and no route to synthesis.
- **The preclinical package.** Neurocrine has not placed the supporting animal work in a peer-reviewed journal or, so far, in a conference abstract that names the compound.
- **Receptor potency ratios.** “Balanced glucagon activity” is described qualitatively. The actual EC50 ratios across the three receptors are the numbers that would let anyone compare it to retatrutide on paper, and they are not public.
- **Trial registration detail.** Coverage refers to a Phase 1 in adults across BMI categories; the granular design has not been laid out publicly at the time of writing.

## Open questions for researchers

For anyone tracking this receptor combination, the questions that will matter over the next twelve months are narrow and answerable:

- Does a deliberately sub-maximal glucagon receptor component change the tolerability profile relative to compounds in the same class, or does it simply shift where the ceiling sits?
- Does the intended pairing with a CRF2 agonist reflect an additive hypothesis about body composition, or a tolerability hedge?
- How does an internally discovered triple agonist entering Phase 1 in 2026 differentiate itself in a field where a same-target molecule is already in Phase 3?
- Will the sequence be disclosed at all before Phase 2, or does Neurocrine hold it as trade secret through the programme?

## Availability and sourceability

NBIP-1968 is not obtainable. It is not catalogued by chemical or peptide suppliers, no CAS number is in circulation, and the structure has not been disclosed — which means there is no synthesis route available to a third party even in principle. Anything offered for sale under this name would not be this compound. Portal to Wellness does not stock it and has no expectation of stocking it.
