Overview
ARA-290 (also known as Cibinetide) is a synthetic 11-amino acid peptide based on the Helix B region of erythropoietin (EPO) — specifically designed to activate the innate repair receptor (IRR, composed of the EPO receptor homodimer or the tissue-protective heteroreceptor with CD131/βc) while avoiding the hematopoietic erythropoietin receptor (EPOR homodimer). This molecular engineering approach has allowed researchers to study EPO’s non-hematopoietic tissue-protective effects — in neurological, metabolic, and inflammatory models — independently of its red blood cell-stimulating activity, providing an important mechanistic tool for dissecting EPO receptor biology.
What It Is
ARA-290 is an 11-amino acid linear peptide derived from the Helix B surface of human EPO, with MW approximately 1,264 Da, CAS number 1234356-69-4. Its sequence corresponds to EPO residues 58-82 (Helix B) and lacks the EPOR homodimer binding epitopes required for erythropoiesis. It preferentially engages the innate repair receptor complex (heterodimer with CD131/βc) at peripheral and central tissue sites, activating cytoprotective signaling cascades including PI3K/Akt and NF-κB without stimulating hematopoiesis.
