What each compound is studied for.

The hallmarks of ageing are a framework from the ageing-biology literature, not a treatment plan. Epithalon is studied in relation to telomerase activity and pineal signalling, largely in animal and early human work from a narrow group of research programmes. Thymalin is a thymic peptide fraction studied for effects on T-lymphocyte differentiation. NAD+ is the co-enzyme the electron transport chain and the sirtuin enzymes depend on, and it declines with age. SS-31 concentrates in the inner mitochondrial membrane and binds cardiolipin. Each is a distinct question at a distinct stage of evidence, and none of it establishes that taking them together does anything in a person.

What’s in this stack.

COMPOUND 01
Epithalon — 40mg/vial

Tetrapeptide (Ala-Glu-Asp-Gly) corresponding to the active site of epithalamin. Studied in relation to telomerase activity and pineal signalling, largely in animal work and early human work from a narrow group of research programmes.

COMPOUND 02
Thymalin — 10mg/vial

Thymic peptide fraction. Studied for effects on T-lymphocyte differentiation. The thymus involutes with age; whether that is modifiable is the question the literature is examining, not a settled finding.

COMPOUND 03
NAD+ — 500mg/vial

Cellular energy co-enzyme. Required for mitochondrial electron transport and the substrate the sirtuin enzymes depend on. Tissue levels decline with age; whether supplementation changes that in a person is a separate and far less settled question.

COMPOUND 04
SS-31 — 50mg/vial

Mitochondria-targeted tetrapeptide. Concentrates in the inner mitochondrial membrane and binds cardiolipin, the phospholipid that scaffolds the respiratory chain. Studied mainly in rare mitochondrial disease populations, which does not generalise to healthy physiology.

What the published work describes.

1

Epitalon — Telomerase and Pineal Signalling

A synthetic tetrapeptide corresponding to the active site of epithalamin, the pineal peptide. Cell and animal work reports effects on telomerase activity and on melatonin secretion patterns. That literature comes largely from one group of research programmes and has not been widely replicated.

2

Thymalin — Thymic Peptide Research

A purified thymic peptide fraction studied for effects on T-lymphocyte differentiation. The Khavinson longevity work reports changes in immune parameters in aging subjects; it is a small and geographically concentrated body of evidence. Nothing here describes an effect in combination with any other compound.

3

NAD+ — Sirtuin and DNA Repair Activation

The primary co-enzyme substrate for the sirtuin deacetylase family (SIRT1-7) and for PARP1/2 in DNA strand-break repair. Tissue NAD+ declines with age. Whether raising it changes anything downstream in a person is the open question the precursor trials are attempting to answer, and those trials are mostly small and short.

4

SS-31 — Mitochondrial Membrane Protection

A tetrapeptide that concentrates in the inner mitochondrial membrane due to its alternating aromatic/cationic structure, where it binds cardiolipin — the phospholipid anchoring cytochrome c and cristae junction proteins. Research examines whether preserving that structure preserves electron transport efficiency under oxidative stress.
