Mechanism of Action
SS-31 (Szeto-Schiller 31) is a tetrapeptide that concentrates in the inner mitochondrial membrane, where it binds cardiolipin — a phospholipid critical for cristae architecture and electron transport chain efficiency. Published work describes stabilisation of cardiolipin and reduced cytochrome c dissociation, with effects on the structural integrity of oxidative phosphorylation complexes. Its mechanism is described as membrane-structural rather than ROS-scavenging.
Research Overview
The MMAD clinical trial evaluated exercise capacity and cardiac function with elamipretide (SS-31) in heart failure patients. Animal studies report restoration of mitochondrial membrane potential and ATP production in aged skeletal muscle, kidney protection in ischaemia-reperfusion injury, and cognitive endpoints in neurodegeneration models. The mechanism studied is structural membrane binding rather than ROS scavenging.
Research & Stack Notes
SS-31 and MOTS-c act through different described mechanisms — membrane structure versus AMPK-mediated metabolic signalling. No adverse interactions with other research compounds are documented in the published literature.

Overview
SS-31 (Szeto-Schiller Peptide 31, also known as Elamipretide or MTP-131) is a synthetic mitochondria-targeted tetrapeptide that selectively concentrates in the inner mitochondrial membrane where it interacts with cardiolipin — the signature phospholipid of the mitochondrial inner membrane. By stabilizing cardiolipin’s interaction with cytochrome c and Complex IV of the electron transport chain, SS-31 has been studied in published research as a tool for examining mitochondrial cristae integrity, electron transport chain efficiency, and mitochondrial reactive oxygen species (mitoROS) generation. Its clinical development as Elamipretide in heart failure and Barth syndrome has produced a substantial published pharmacology literature.
What It Is
SS-31 is a synthetic tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH₂ (Dmt = 2’6′-dimethyltyrosine), MW approximately 639 Da, CAS number 736992-21-5. The alternating aromatic and basic amino acid residues produce a net positive charge that drives electrostatic accumulation in the inner mitochondrial membrane, concentrating the peptide ~1000-fold relative to cytoplasm without dependence on mitochondrial membrane potential. The Dmt residue’s electron-rich phenol ring is responsible for cardiolipin interaction and the peptide’s radical-scavenging activity.

Regulatory update — August 2026: Elamipretide received FDA accelerated approval as Forzinity (elamipretide HCl) on September 19, 2025, for a single narrow indication: improving muscle strength in Barth syndrome patients weighing at least 30kg. This is the first mitochondria-targeted therapy to reach FDA approval. That approval does not extend to longevity, general mitochondrial support, insulin sensitivity, or any other use. The SS-31 sold on this page is a separate, unaffiliated research-use product — it carries no FDA approval of its own. See our mitochondrial peptide comparison for more detail.
