Overview
IGF-1 LR3 (Long-Arginine-3 IGF-1) is a synthetic long-acting analogue of human insulin-like growth factor 1 (IGF-1) engineered for research applications requiring extended receptor activation and reduced IGFBP binding. By substituting glutamate at position 3 with arginine and prepending a 13-amino acid extension peptide at the N-terminus, IGF-1 LR3 achieves markedly reduced binding to IGF-binding proteins (IGFBPs) and a correspondingly extended plasma half-life compared to native IGF-1 — making it the preferred research tool for sustained IGF-1 receptor pathway studies.
What It Is
IGF-1 LR3 is an 83-amino acid synthetic analogue of human IGF-1, incorporating a 13-residue N-terminal extension and a Glu³→Arg substitution. Its molecular weight is approximately 9,117 Da. It has ~2–3% of the IGFBP binding affinity of native IGF-1, resulting in an estimated half-life of 20–30 hours in vivo versus the ~12-minute half-life of native IGF-1. CAS number: 946870-92-4.

Frequently Asked Questions
IGF-1 LR3 is a modified analogue of insulin-like growth factor 1 with an N-terminal extension and an arginine substitution that reduce binding to IGF-binding proteins, giving it a longer half-life than native IGF-1. It is widely used as an IGF-1 receptor agonist in cell-culture and growth-factor signalling research.
