- Studied for BDNF and NGF gene expression in hippocampus and prefrontal cortex models
- Developed at the Russian Academy of Sciences; studied in ischaemic stroke recovery
- No stimulant mechanism or receptor downregulation documented in the published literature
- Derived from the ACTH(4–7) sequence with a C-terminal Pro-Gly-Pro extension for metabolic stability
- No receptor competition with other research peptides documented
Mechanism of Action
Semax is a heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the ACTH(4–7) sequence, modified with a proline-glycine-proline C-terminal extension for metabolic stability. Published work reports upregulation of BDNF and NGF gene expression in the hippocampus and prefrontal cortex, effects on dopaminergic and serotonergic neurotransmission, and neuroprotective effects against ischaemic and oxidative stress. The Pro-Gly-Pro extension confers resistance to enzymatic degradation.
Research Overview
Clinical studies in Russia evaluated Semax in acute ischaemic stroke recovery, reporting changes in neurological deficit scores versus placebo. Cognitive studies report effects on attention, working memory and processing speed. BDNF upregulation has been observed within hours of administration, with expression reported to persist for several days. This body of work has limited independent replication outside Russia.
Research & Stack Notes
Semax and Selank are frequently examined together in the published cognitive literature, acting on different pathways. No receptor competition with longevity or metabolic research compounds is documented.

Overview
Semax is a synthetic nootropic heptapeptide developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, based on the ACTH(4-10) core fragment extended with a Pro-Gly-Pro C-terminal tripeptide for improved metabolic stability. Originally studied for cerebrovascular and neuroprotective applications, Semax has generated a published research base spanning BDNF upregulation, dopaminergic and serotonergic system modulation, ischemia-reperfusion research, and cognitive function biomarker studies in rodent models. It holds regulatory approval in Russia and Ukraine as a nootropic peptide drug.
What It Is
Semax is a heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro, MW approximately 779 Da, CAS number 80714-61-0. It is a structural analogue of ACTH(4-7) (Met-Glu-His-Phe) with the C-terminal Pro-Gly-Pro extension borrowed from the Selank research program to confer enzymatic stability. Semax does not bind melanocortin receptors at pharmacologically relevant concentrations but has been shown in published research to upregulate BDNF and NGF expression, modulate dopamine system markers, and affect serotonin transporter activity.
Key Research Applications
- BDNF and NGF upregulation in cortical and hippocampal tissue research
- Cerebrovascular research — ischemia-reperfusion injury and neuroprotection modeling
- Dopaminergic system modulation and D1/D2 receptor expression studies
- Serotonin transporter (SERT) and serotonergic signaling research
- Cognitive function biomarker and memory consolidation studies in rodent models
- ACTH fragment pharmacology without adrenocortical activation
- Nootropic peptide comparative pharmacology vs. Selank and other ACTH-derived peptides
- Stroke, traumatic brain injury, and hypoxia preclinical modeling studies

Regulatory Status Note
On July 23–24, 2026, FDA’s Pharmacy Compounding Advisory Committee reviewed whether BPC-157, TB-500, KPV, MOTS-c, Semax, Epitalon, and emideltide (DSIP) should be added to the 503A Bulks List for pharmacy compounding. The committee voted to recommend six of the seven; emideltide (DSIP) was not recommended. FDA’s own briefing documents had recommended against all seven.
A committee vote is a recommendation only. It does not constitute FDA approval, does not establish safety or effectiveness, and does not change the status of this product, which is sold strictly for research use. FDA must complete formal rulemaking before any substance is added to the list. Source: FDA Advisory Committee Calendar. Last reviewed: August 17, 2026.
