Overview
Thymosin Alpha-1 (Tα1) is a 28-amino acid peptide originally isolated from thymic tissue and subsequently characterized as a potent immunomodulatory agent with a broad published research base spanning innate and adaptive immune system biology, antiviral research, and cancer immunology. First identified by Allan Goldstein’s research group at George Washington University as the biologically active component of the thymus-derived fraction Thymosin Fraction 5, Tα1 has been studied in published literature across multiple continents and holds regulatory approval as a prescription pharmaceutical (Zadaxin) in several countries for hepatitis and immune-deficiency applications.
What It Is
Thymosin Alpha-1 (Tα1) is a 28-amino acid N-acetylated peptide with molecular formula C₁₂₉H₂₁₅N₃₃O₅₅ (acetylated form), MW approximately 3,108 Da, CAS number 62304-98-7. The N-terminal acetylation is essential for biological activity. Tα1 acts on multiple immune cell populations through pattern recognition receptor pathways including TLR signaling, TRIF-dependent interferon regulatory factor activation, and dendritic cell maturation pathways, resulting in downstream NK cell, cytotoxic T-cell, and Th1 immune response enhancement.
Key Research Applications
- Thymic peptide immunomodulation and T-cell differentiation/maturation research
- TLR2, TLR7, TLR9 signaling and innate immune activation pathway studies
- NK cell and cytotoxic T-lymphocyte (CTL) activity enhancement research
- Dendritic cell maturation and antigen presentation research
- Th1/Th2 immune response polarization and cytokine profile studies
- Antiviral research — interferon production and viral replication suppression models
- Cancer immunology — tumor microenvironment immune cell activity research
- Sepsis and immune exhaustion recovery modeling in animal systems
