Overview
Sirolimus (rapamycin) is one of the most significant compounds in modern longevity and cellular biology research. Originally isolated from Streptomyces hygroscopicus bacteria discovered on Easter Island (Rapa Nui), sirolimus is the prototypical mTOR (mechanistic target of rapamycin) pathway inhibitor. The mTOR signaling node integrates cellular nutrient sensing, growth factor signals, and energy status to regulate protein synthesis, autophagy, and cellular aging programs. Rapamycin’s ability to extend lifespan in multiple model organisms — including mice — has made it the reference compound in translational longevity biology.
What It Is
Sirolimus is a macrolide compound (molecular formula C₅₁H₇₉NO₁₃, MW ~914.17 Da, CAS number 53123-88-9) that acts as an allosteric inhibitor of mTORC1 by forming a gain-of-function complex with the FK506-binding protein FKBP12. This FKBP12-rapamycin complex then binds and inhibits mTOR’s kinase activity toward S6K1 and 4E-BP1. Sirolimus is FDA-approved as an immunosuppressant (Rapamune), providing a substantial human clinical pharmacokinetics literature.
